Does Glyde appear in genocide?
Glyde is a Special Encounter, which appears on Wave 5 of Genocide Snowdin with a 20% chance.
This encounter must happen before the fight with Papyrus, or Glyde does not appear. The higher the kill count of the protagonist during the Genocide Route, the longer it takes for Glyde to appear — possibly upwards of thirty minutes.
If you kill a secret boss in a pacifist run, will it be a neutral run then? Yes, killing any monsters will lock you out of a pacifist run and revert it back to a neutral one. This includes the hidden Kickstarter-backer enemies Glyde and So Sorry.
The Genocide Route leads to one of the three main endings of Undertale. It consists of the protagonist killing every possible monster in the Underground. This route's ending can occur without prior completion of the game.

Killing every creature and also purposefully triggering every random encounter to kill those creatures as well will get you the genocide ending. Doing this will permanently taint your save file, even for future playthroughs.
Destroying the Dummy does not abort the attempt at the True Pacifist Route and neither talking, nor fleeing, nor boring the Dummy aborts the Genocide Route attempt. If the protagonist talks to the Dummy, Toriel congratulates them.
In order to encounter Glyde, the protagonist must go to the Mysterious Door room outside Snowdin (with the four mushrooms), and move for at least two minutes. This encounter must happen before the fight with Papyrus and if the protagonist did not kill any fleeable monster in Snowdin, or Glyde does not appear.
He doesn't yield any exp IIRC, so he can just be ignored. Jerry yields 1 EXP, so killing him will prevent you from earning a Pacifist runthrough.
Can you spare sans in a Genocide run?
You can't spare Sans in game, although you can skip past his bossfight, which, in a way, is sparing sans because you aren't killing him.
You will need to have murdered 20 creatures. After you've met the quota the next random encounter will look like this, with the text "But nobody came." Head back to the branching room and exit north where you'll reunite with Toriel who will take you to her home.
No matter what ending you get in Undertale, the Deltarune universe will be the same. All endings of Undertale are canon.
So Sorry is a hidden miniboss the protagonist encounters in the Art Club Room. He was a bought-in character during the Kickstarter for the game.

In order to Spare him you'll need to Beckon him, Pet him, Play with him, then Pet him twice more. Once he's defeated, head east for Papyrus's last puzzle which is even less successful than his previous attempts.
Gaster is a hidden boss who unlike his Final counterpart, is located in the True Lab.
Temmie is a secret monster that can only be obtained by defeating it in the genocide version of waterfall. She has a 20% chance of spawning at the end of wave 4; upon being defeated, you will obtain Temmie.
And this will have spoilers, so if you have not played or seen the game, I advise you to leave. Now Sans is the hardest boss in the whole game. You fight him when you do a genocide run, and he inflicts poison damage when you get hit. And there are no invincibility frames, you will continue to take damage.
What happens if you name your Undertale character Frisk? Naming the fallen child 'Frisk' enables hard mode, however the run ends after the boss fight with Toriel as the developer, Toby Fox, never got around to finishing hard mode.
What happens if you do Genocide twice?
The only changes that occur after performing a Genocide route is the change in the True Pacifist post-credit cutscenes, and Chara's dialogue at the end of another Genocide route. Neutral routes are the same as before.
Soulless Pacifist occurs when attempt to do the True Pacifist run but you have already completed the Genocide run. The only changes are in the very last scene after you chose to either stay with Toriel or go somewhere else. While lying in their bed Frisk wakes up and makes the =) face with their eyes being red.

In the Genocide Route, Glad Dummy can be killed or spared immediately. Performing other actions (missing attacks, checking, or using items) for eight consecutive turns causes Glad Dummy to get bored and float away. Sparing or boring Glad Dummy aborts the Genocide Route.
If you play the genocide ending you will never be allowed to get the true pacifist ending on that copy/account ever again. Each time you do a pacifist run after the first time you do genocide the game will end with a sinister cutscene implying you'll kill everyone anyway.
